Clotrimazole is a broad-spectrum antifungal agent that inhibits the growth of pathogenic yeasts by changing the permeability of cell membranes. The action of clotrimazole is fungistatic up to 20 mcg/mL and may be fungicidal in vitro against Candida species. Resistance, once rare, is now more common in many patient groups.
Clotrimazole acts primarily by damaging the permeability barrier in the fungal cell membrane. It causes inhibition of ergosterol biosynthesis, an essential component of fungal membranes. Lack of ergosterol prevents the formation of functional cell membranes, leading to dose-dependent inhibition of fungal growth.
